چکیده

Thein vitro s tudy of the combined effects of humoral antibody and lymphocytes has been essential in attempting to understand the paradoxical effect of humoral antibodies leading to the phenomenon of enhancement (1). Par t of the mechanism has been considered to be the shielding of cellular antigenic determinants by humoral antibody from the lymphocytes. This could mask antigenic sites preventing recognition and result in an afferent block and also interfere with the cytotoxic effect of specifically sensitized lymphocytes producing an efferent block. Support for both these hypotheses has been demonstrated (2-7). The blocking effect of humoral antibodies has been proposed to be of importance in the phenomena of tolerance, chimerism, and the protection of the fetus from maternal lymphocytes (8). However, it has been well documented in certain systems that humoral antibody and lymphocytes may act in concert to effect target cells' destruction (9, 10). Under different experimental conditions, pretreatment of target cells with humoral antibody may protect them from the cytotoxic effect of immune lymphocytes or mediate their destruction by a normal lymphocyte population. In vivo probably a balance between these two phenomena occurs. Enhancement of normal lymphocyte cytotoxicity by sera with high antibody titers against H-2 or virus-associated antigens.

The experiments reported in this paper were designed to investigate the in vitro interaction of immune sera with different antibody titers as determined by indirect mem brane immunofluorescence (MF) with normal or immune lymphoid cells in mediating target cell destruction. Target cells in these experiments were two Moloney sarcoma virus (MSV)-induced tumor cell lines derived from BALB/c and C57...

In vitro induction of T-lymphocyte-mediated cytotoxicity by infectious murine type C oncornaviruses

We have developed a system to induce oncornavirus-specific secondary cytotoxic response in vitro. When Moloney strain of murine sarcoma virus-immune spleen cells were cultivated with purified infectious Moloney murine leukemia virus (M-MuLV) or with supernates of tissue culture cells containing infectious virus, a virus-specific secondary cytotoxic response directed against type-specific determ...

Induction of Moloney murine sarcoma virus tolerance in adult mice by anti-CD4 monoclonal antibody treatment.

The role of CD4+ cells in the immune response to Moloney murine sarcoma/leukemia virus complex was studied in adult mice undergoing long-term CD4+ cell functional depletion by treatment with anti-CD4 H129.19 monoclonal antibody (immunoglobulin G2a, kappa). Adult mice given injections of Moloney murine sarcoma/leukemia virus complex 1 day or 2 wk after monoclonal antibody treatment died with pro...

The Cytotoxic and Synergistic Effects of Flavonoid Derivatives on Doxorubicin Cytotoxicity in Hela, MDA-MB-231, and HT-29 Cancer Cells

Background: Flavonoids have a variety of biological activities, such as anti-allergic, anti-inflammatory, anti-oxidative, free radical scavenging, and anti-mutagenic. Methods: The cytotoxic effects of three synthesized flavonoid derivatives (K3, K4 and K5) were evaluated against Hela , MDA-MB -231 and HT-29 cancer cells using MTT assay. Results: The results showed that these flavonoids were n...

Antibody-dependent cell-mediated cytotoxicity in the Moloney sarcoma virus system: differential activity of IgG and IgM with different subpopulations of lymphocytes

Antibody-dependent cell-mediated cytotoxicity in the Moloney sarcoma virus (MSV) system was evaluated in terms of the differential ability of IgG and IgM from MSV regressor animals to induce cytotoxicity by lymphocytes from lymph node, spleen, and thymus. The cell-mediated cytotoxicity induced by both IgM and IgG was specific for target possessing the appropriate virally determined cell surface...
